'Pacific Rim' WonderCon Footage Destroys All Monsters

When the first trailer for "Pacific Rim" hit, there were essentially two kinds of reactions. There was the response from those familiar with Guillermo del Toro's work in movies like "Hellboy" and "Pan's Labyrith" who were looking forward to a new movie from the Mexican director after a number of false starts. Then there were [...]

Sanford deflects talk of affair in South Carolina debate

By Harriet McLeod

CHARLESTON, South Carolina (Reuters) - Former Governor Mark Sanford deflected criticism of an extramarital affair leveled in a debate on Monday by his opponent for a South Carolina congressional seat, political newcomer Elizabeth Colbert Busch.

Sanford, a Republican, is hoping to revive his political career after he famously disappeared from the state in 2009 while he was governor to visit his mistress in Argentina, telling aides that he was hiking the Appalachian Trail.

Colbert Busch, the sister of television comedian and political satirist Stephen Colbert, is a Democratic businesswoman making her first run for public office.

The election to fill the seat left vacant when Republican congressman Tim Scott was appointed to the Senate will be held on May 7.

Sanford formerly held the congressional seat between 1995 and 2001. In addition to the affair, he has been hurt by accusations by his ex-wife, Jenny Sanford, that he trespassed on her property.

"Some in the national media seem to think it's a race between Jenny Sanford and Stephen Colbert," moderator John Avlon said in opening the debate.

For the most part, the candidates sought to prove otherwise.

While they spent much of the debate carving out their differences on federal spending and immigration, the debate's most peculiar moment came when Colbert Busch raised the Appalachian Trail incident but failed to get a reaction out of Sanford, who said he had not heard the jab.

"When we talk about fiscal spending and we talk about protecting the taxpayers, it doesn't mean you take that money we saved and leave the country for a personal purpose," Colbert Busch said.

Amid boos and laughter from the audience, Sanford said: "I couldn't hear what she said."

"Repeat it, I didn't hear it, I'm sorry," he said.

COLBERT BUSCH LEADS POLL

A Public Policy Polling survey released earlier this month found Colbert Busch leading among likely voters 50 percent to Sanford's 41 percent.

The poll followed the revelation that Sanford's ex-wife had accused him in court documents of trespassing at her home, and the National Republican Congressional Committee's announcement it would no longer take part in Sanford's campaign.

Sanford said he had gone to his ex-wife's house in February while she was out of town to watch the Super Bowl with one of their sons and that he had been unable to reach her to ask for the permission required under their divorce settlement.

For much of the debate, Sanford sought to remind voters of his conservative credentials, while Colbert Busch sought to dismiss Sanford's claim that, if elected, she would do the bidding of labor unions and her liberal supporters in Congress.

"I want to be very clear, Mark," Colbert Busch said. "Nobody tells me what to do. I am a fiscally conservative, independent tough businesswoman. Let's talk about reaching across the aisles and being practical."

Colbert Busch, business development director of Clemson University's Restoration Institute, said that, if elected, she would cut her own salary by $10,000.

Sanford touted his record of fiscal conservatism in three terms in Congress and two terms as governor.

"I would look back on my time in Congress as a short patch in Washington when budgets were actually balanced. I was against earmarks before being against earmarks was cool," he said.

On immigration reform, Sanford argued for a more robust guest worker program and an extended "low-tech fence."

Colbert Busch said she supports legislation in the U.S. Senate that offers a "tough and fair" path to citizenship for illegal immigrants.

While Colbert Busch's famous brother did not come up, Sanford did get one other opportunity to address the scandal that derailed his political career.

Asked if he regretted voting for President Bill Clinton's impeachment in 1998, after Clinton admitted to an affair, Sanford responded by saying he would reframe the question.

"Do you think that President Clinton should be condemned for a mistake he made in his life for the rest of his life?" Sanford said.

Marissa Mayer Explains Yahoo's New Approach ... - Business Insider

Owen Thomas, Business Insider

Yahoo CEO Marissa Mayer at the 2013 Goldman Sachs Technology and Internet Conference

?

Yahoo CEO Marissa Mayer has a new plan for generating more ad revenue.

Today she announced two new types of ads: Yahoo! Stream Ads and a new interactive Billboard ad.

This is all part of the big redesign that Yahoo launched in February, Mayer says. That's when Yahoo changed its front page into a streaming headline format, eliminating several ad units and letting people customize the page in new ways.

Today she's added some ad units back to the page. As its name implies the Stream ad will be embedded into the news stream of a user's home page. It will show up across devices like desktops/laptops, smartphones, and tablets.

It looks like this:

Yahoo

Yahoo Stream ad is the text in the yellow box

The Billboard unit will look like a fairly traditional banner ad that sits on top of the Yahoo page. Mayer says that it is different because it will ink to content that's more "fun and engaging" for Yahoo visitors.

The first Stream ads could start showing up as soon as this week.

Here's her full blog post:

As we continue to build products and features that inspire and entertain our users, we're committed to delivering engaging and effective advertising opportunities. Over the past few months, we?ve begun to evolve the Yahoo! experience to be more personal, intuitive, and immersive -- incorporating more modern paradigms like our news stream. Today, we?re matching that engaging stream experience with a new advertising format -- Yahoo! Stream Ads.

Since we launched our Yahoo! news stream in February, our users have responded by visiting more, staying longer, and increasing their engagement with our content. Like with web search, users appreciate complementary, unobtrusive advertising, and we?re committed to delivering just that. Advertising can, and should, enhance content discovery in a seamless and effective manner. The more our users spend time with Yahoo!, the more relevant and personalized the content and advertising becomes. Stream Ads are the sponsored twin to our newsfeed articles and are every bit as personalized and engaging.

Today, we?re also announcing a new Yahoo.com Billboard designed to deliver richer content interactions to Yahoo! users. For example, a movie trailer that runs on the Billboard could link to more information about the film and cast, and let you buy tickets directly from the ad. We believe that advertising like this can be fun and engaging for users, while also effective for advertisers.

Going forward, we?re committed to advertising formats that complement our products and content, and enhance the user experience. Stay tuned for more in the coming months!

10 Event Ideas to Boost Your Blog Traffic | SiteProNews: Webmaster ...

In the business world, an event is a public gathering for the purpose of networking and/or marketing.? Holding events on your blog or website can help give your traffic a substantial boost.? By holding different events, you bring more life and interaction to your blog that, in turn, will bring more traffic.

Contests and giveaways are effective events to stir up some interest and excitement.? If you are not sure which type of event to consider, below are 10 ideas to help you to get started.

1. Charity Week ? Offer to pay a percentage of all orders to charity.? Have your visitors post information about their favorite charities and donate to them.? Present prizes, discounts, gift cards, etc., for your visitors.? Hold auctions with your products to raise money for various charities.? Ask people to help you raise money by making others aware of your charity event. ?NOTE: Please be sincere in your efforts to raise money for charity.? Do not just use it as a marketing ploy.

2. Meet-and-Greet Event ? Hold a meet-and-greet event for other bloggers.? Invite bloggers to come to your event to meet, network, and brainstorm with fellow bloggers.? You organize a couple of games for prizes ? trivia games are always popular.? Host a question-and-answer forum to offer advice, experience and expertise.? Present guest speakers to share their knowledge and expertise as well.

3. Comment Contest ? Let your readers know that if they post a comment, (with substance, not just thank you for the info or anything similar) they will have a chance at winning a prize.? You can do a random draw for the prize or you could have a secret word (which directly relates to your content) and anyone who uses that secret word in their comment gets a shot at the prize.? Make it fun and use different tactics each time.

4. Holiday Events ? Host a costume contest on Halloween.? Have people post pictures of themselves in their costumes and then have all posters vote for their favorites.? The one with the most votes wins.? For Christmas, you could have a ?Find Santa? contest.? Put a picture of Santa somewhere on your site and the first person to find it wins. ?For Easter, you could have an Easter egg hunt. The possibilities are endless.

5. Guess the Location Contest ? Post a picture of a semi-famous location (don?t make it too easy) along with some vague clues and challenge your readers to guess the site.? Offer a new clue each day until someone discovers the location in the photo.

6. Photo Contest ? Ask your visitors to post a photo they took along with their comment and let everyone vote for the best picture.? People love to vote on things and they love to win prizes, so it is a win-win contest!

7. Parade of Blogs ? Does your community ever hold a Parade of Homes?? You could do something similar with blogs.? Contact other blog owners and see if they would be interested in participating.? You again could let people vote on the blogs they liked best for certain categories such as products, content, user-friendliness, etc.??This would be a great way for blog owners to learn how to make their blogs more successful. It is also a good opportunity for networking.

8. Group Discussions ? Hold monthly group discussions similar to a book club only you could discuss ways to increase sales, traffic or build your business in general.

9. Trivia Parties ? There aren?t too many people who don?t enjoy a good trivia challenge so why not hold periodical trivia parties.

10. Scavenger Hunt ? Have a list of objects for people to find on the Internet along with clues on where to can find them.?Give participants in which to find the objects.?Ask them give you the URL from the site where they found the object so you know it is the right one.?Just doing a Google search for each object and going to any site is not allowed!? Make it a specific object on a specific site.

Proper planning will be a big part of your event, so don?t try to schedule these events on the spur of the moment.? Be precise and effective in your planning so all goes well.

I hope these ideas help to inspire you to organize your own successful event.? Hold events and contests on a regular basis to keep your blog active, alive and profitable.

Don?t be one of the 95 percent of people who fail at their online business.? Terri Seymour can help you make money online.? Find out how to increase your traffic and sales with her popular How to Build Your Online Business eBook for Free at:?http://www.SeymourProducts.com.

Chrysler revenue slumps on new model launches

By Bernie Woodall and Jennifer Clark

DETROIT/MILAN (Reuters) - Carmaker Fiat's first-quarter profit slumped more than expected as its U.S. unit Chrysler's sales suffered from the phase-out of the Jeep Liberty pending a new model launch.

"We knew we would be limping in the quarter. I just didn't think I was going to limp that much," said Sergio Marchionne, chief executive of both Chrysler and its parent Fiat, on a conference call with analysts and reporters.

Marchionne in January warned that Chrysler's first-quarter earnings would be down from a year earlier because of the expense of product launches and the fact that the Jeep Liberty SUV was no longer being produced.

The Liberty's successor, the Jeep Cherokee, was not sold in the first quarter and will not be sold until the third quarter.

But Marchionne said that there were delays in the production launches of the Jeep Grand Cherokee and RAM heavy duty trucks, which he said are among the highest profit-makers.

Fiat, which took control of the third-largest U.S. automaker when Chrysler emerged from a government-sponsored bankruptcy four years ago, stood by its financial forecasts for 2013 despite a worsening European car market and lower revenue in the U.S. for the first quarter.

Fiat sees full-year revenue in the 88 billion euro to 92 billion euro range, trading profit between 4 billion and 4.5 billion euros, and net industrial debt of about 7 billion euros.

Analysts had expected a weak quarter for the combined group, and the results came in below consensus.

Trading profit was 618 million euros, below the 720 million euros forecast by 21 analysts published on Fiat's website. Pre-tax profit came in at 160 million euros, compared to a forecast of 300 million euros by analysts.

At Chrysler, first-quarter net income fell 65 percent to $166 million from $473 million a year earlier. Net revenue slipped 6 percent to $15.4 billion.

Net debt was 7.10 billion euros, in line with forecasts, and higher than the 6.5 billion euros at the end of 2012.

Citi Research analyst Harald Hendrikse said the quarter's results again highlight Fiat's difficult debt issue.

"With debt at current levels, the company cannot compete in this industry longer term against giants like Volkswagen AG spending 16 billion euros on capital expenditures and research and development," said Citi in a note.

NARROWING EUROPEAN LOSSES

Fiat's European losses before interest and tax narrowed to 111 million euros, from a year-ago loss of 170 million euros, despite a 10 percent drop in industry sales in the region. Marchionne said he expects Fiat to narrow its operating losses in Europe further in 2013.

Fiat outperformed a dismal European car market with the launch of the new Fiat 500L small car, the company said.

Fiat, like competitors Ford , General Motors and Peugeot , is losing money in Europe as buyers delay purchases of new cars until the economy improves. Unlike rivals, however, Fiat has decided to invest in retooling its idled factories to ramp up volumes of higher-margin cars for export rather than close plants.

The company aims to break even in Europe by 2015 or 2016.

LOOKING AHEAD

Chrysler said it will increase vehicle shipments in the second quarter by at least 13 percent from the first quarter, to 650,000 vehicles up from 574,000 in the first quarter.

Marchionne said the Jeep Grand Cherokee, now that its launch delays are over, will show strong April sales, and that the biggest boost in Grand Cherokee sales will show in the third quarter.

Marchionne said in the coming quarters Chrysler will better match up production and retail sales performance.

"Close your eyes, plug your nose and move it on from here," said Marchionne to analysts on a conference call.

COMING TO AMERICA?

Looking further out into the future, Marchionne said he hopes to merge Fiat and Chrysler into one group and he hinted strongly that the combined company will be headquartered outside of its native Italy.

Many observers expect the merged company to be headquartered where Chrysler is now, in Auburn Hills near Detroit, and to be listed on the New York Stock Exchange and in Italy.

Marchionne said the merged company would be headquartered in the geographical region that has "the adequacy of capital markets (necessary to) support our operations going forward."

Marchionne said that "Europe is becoming a less and less relevant fact in the scheme of things" as its share of the global auto market diminishes.

Italy has been a particularly weak player in a weakening region for auto sales.

"Italy in 2012 represented 10 percent of the overall sales of this (Fiat) Group," aid Marchionne. "And I think it's a stark reality for someone who has been a Fiat aficionado all his life. This is a different house. It looks at the world in a completely different way."

Egypt president, judges compromising on reform law

CAIRO (AP) ? The Egyptian president's office indicated Sunday a compromise has been reached with the judiciary to defuse an uproar over a proposed law that would have forced out thousands of the country's most senior judges.

Just three days earlier, the country's Islamist-led parliament pushed ahead with the disputed bill that would have lowered the retirement age for judges from 70 to 60. That would affect nearly a quarter of Egypt's 13,000 judges and prosecution officials.

The draft also would have barred the courts from reviewing or overturning presidential decrees issued by the president late last year, including his unilateral appointment of a new top prosecutor.

The crisis over the judiciary is a reflection of the deep polarization that has split the country over the rule of Morsi and his Muslim Brotherhood party. Morsi has been in a power struggle with the judiciary since his June election.

In an attempt to resolve the situation, Morsi met Sunday with five top judges. A statement from the president's office after the meeting said Morsi will launch a conference this week to work out a compromise with judges regarding laws that affect the judiciary.

"The president said he will personally adopt all the conclusions of the conference for proposals of bills in order to submit them to the legislative council," the statement said.

After the meeting, opponents of the proposed law canceled protests set for Monday.

The president's allies say the courts are filled with loyalists of the deposed regime of Hosni Mubarak. The opposition accuses Morsi's backers of calling for reform of the judiciary as a cover to install their own supporters.

A protest by opponents and supporters of the bill led to street clashes in the capital nine days ago, and thousands of judges met last week to demand international organizations investigate the crisis. The dispute also prompted the resignations of top aides and advisers to the presidency.

The announcement by the president's office effectively shelves the law that was under discussion by lawmakers. A new proposal for a judicial reform law will likely first be agreed upon by Morsi and judges, and then be sent to the parliament.

Morsi is also facing divisions within the country's battered police force.

Dozens of Egyptian police officers disobeyed orders Sunday and stormed a superior's office in the capital, shut down a security directorate in the north and went on strike in the south.

It was the year's third wave of strikes by police, who demand incentives like better wages, greater firepower and more benefits.

The police force has not recovered from the days of the 2011 uprising that deposed Mubarak. His police were a symbol of the regime's unchecked powers and abuses, and they were forced from the streets in the early stages of the revolt by angry protesters.

The Interior Ministry, which oversees police in Egypt, relies on low-ranking police to protect government buildings. Hundreds of policemen have been wounded this year and several have been killed in anti-government protests.

Dozens of low-ranking officers stormed the office of the deputy interior minister in charge of health care for police. His office is inside the main police hospital in Cairo's Nileside neighborhood of Agouza. They said promises of better health care have gone unfulfilled.

In the south, police at five stations in the province of Assuit went on strike, charging that the government did not fulfill any of their demands.

In the Nile Delta province of Kafr el-Sheikh, police locked the gate to the security division with chains, according to security officials and state media reports.

Strikes also spread north of Cairo to Sharqiya and the Suez Canal city of Ismailiya.

Some are also calling for the dismissal of Interior Minister Mohammed Ibrahim. The minister met with some of the protesting policemen Sunday.

Others are protesting alleged attempts by the powerful Muslim Brotherhood to try to control the force, a charge the Islamist group denies. For decades, Egypt's police aggressively targeted the Brotherhood, and Morsi himself was imprisoned under Mubarak.

There are concerns that a recent decision to purchase 100,000 new 9mm pistols for police could lead to an even greater use of excessive force against unarmed protesters and civilians.

Officer Mohammed Mustafa was among those who stormed the deputy minister's office in Cairo on Sunday. He told The Associated Press that the group ended its sit-in after superiors vowed to look into their demands.

Though his salary has increased by almost three-fold following the uprising, Mustafa said he still earns only $185 a month. After 15 years on the job, low-ranking officers receive just $2,000 in compensation when they retire.

"Morsi, before he was president, promised that he would solve the problems of the police force," he said. "We want action, not words."

Dream Home win came just in time for growing family ...

VIRGINIA BEACH

Julio and Justine Gavino found their happily ever after thanks to some spur-of-the-moment spending.

The couple had already discussed and bought three tickets for a chance to win the St. Jude Dream Home in Virginia Beach's Ashville Park. However, after an impromptu visit to the house, Justine bought two more $100 tickets.

"I didn't know she was going to buy more tickets," Julio said. "I am glad she did."

The couple, who have 9-month-old twins, learned Sunday that one of the tickets Justine bought at the last minute was the winner. They're now the new owners of a 3,100-square-foot, four-bedroom, 2-1/2-bath home constructed by Stephen Alexander Homes & Neighborhoods and valued at $525,000.

"We were getting ready to go to a birthday party, and I was watching the (WTKR NewsChannel 3 St. Jude) telethon," Julio said. "They pulled out the winning ticket, and Barbara Ciara said the winner lived in Suffolk. I started screaming for my wife to come to watch the TV."

Julio couldn't believe it when his wife's name was called out. He had to rewind the broadcast and watch it again. A few minutes later, they got the call informing them.

"We were both screaming, and we scared the babies, and then they started crying," Julio said. "It was a crazy moment."

The 13th annual St. Jude Dream Home Giveaway set a new record with 12,000 tickets sold and raised $1,229,355, the greatest amount of monies raised in the local campaign's history.

In addition, 23 other prizes, each valued at $1,000 or more, were awarded to people who purchased tickets.

All proceeds, after costs, benefit St. Jude Children's Research Hospital in Memphis, Tenn.

Since the St. Jude Dream Home Giveaway program launched in 1991, St. Jude has given away more than 300 houses and raised more than $240 million in ticket sales, making it the largest single-event fundraiser for the hospital. It's because of programs like this that no family ever pays St. Jude for anything.

St. Jude covers the cost of treatment, travel, lodging and food for patients and a family member.

"I never thought we would really win, but that was OK since the money was going to St. Jude," Julio said. "We are committed to St. Jude mission and really believe in the work that the hospital does."

Julio, an IT network analyst, and Justine, a commercial property consultant, live in Suffolk. They'd already been looking to sell their 1,800-square-foot home and find something bigger in Virginia Beach, where the grew up.

"We wanted to get back into the Ocean Lakes area and be near our family," Julio said. "When I looked up the St. Jude house and saw it was in that area, I told my wife it might be a sign."

Even after buying the tickets for a chance to win the house, the Gavinos still kept looking and planning their return to the Beach. Now, instead of house hunting, they're planning a move.

"The day we won the house was the first day I saw it," Julio said. "We made the decision right then and there that we wanted to move in it. We think God gave us the house, and there's no way we are going to get rid of it. It's the perfect ending to what we have been working for our whole lives."

Scientists create novel approach to find RNAs involved in long-term memory storage

Scientists create novel approach to find RNAs involved in long-term memory storage

Friday, April 26, 2013

Despite decades of research, relatively little is known about the identity of RNA molecules that are transported as part of the molecular process underpinning learning and memory.

Now, working together, scientists from the Florida campus of The Scripps Research Institute (TSRI), Columbia University and the University of Florida, Gainesville, have developed a novel strategy for isolating and characterizing a substantial number of RNAs transported from the cell-body of neuron (nerve cell) to the synapse, the small gap separating neurons that enables cell to cell communication.

Using this new method, the scientists were able to identify nearly 6,000 transcripts (RNA sequences) from the genome of Aplysia, a sea slug widely used in scientific investigation.

The scientists' target is known as the synaptic transcriptome?roughly the complete set of RNA molecules transported from the neuronal cell body to the synapse.

In the study, published recently in the journal Proceedings of the National Academy of Sciences, the scientists focused on the RNA transport complexes that interact with the molecular motor kinesin; kinesin proteins move along filaments known as microtubules in the cell and carry various gene products during the early stage of memory storage.

While neurons use active transport mechanisms such as kinesin to deliver RNA cargos to synapses, once they arrive at their synaptic destination that service stops and is taken over by other, more localized mechanisms?in much the same way that a traveler's bags gets handed off to the hotel doorman once the taxi has dropped them at the entrance.

The scientists identified thousands of these unique sequences of both coding and noncoding RNAs. As it turned out, several of these RNAs play key roles in the maintenance of synaptic function and growth.

The scientists also uncovered several antisense RNAs (paired duplicates that can inhibit gene expression), although what their function at the synapse might be remains unknown.

"Our analyses suggest that the transported RNAs are surprisingly diverse," said Sathya Puthanveettil, a TSRI assistant professor who designed the study. "It also brings up an important question of why so many different RNAs are transported to synapses. One reason may be that they are stored there to be used later to help maintain long-term memories."

The team's new approach offers the advantage of avoiding the dissection of neuronal processes to identify synaptically localized RNAs by focusing on transport complexes instead, Puthanveettil said. This new approach should help in better understanding changes in localized RNAs and their role in local translation as molecular substrates, not only in memory storage, but also in a variety of other physiological conditions, including development.

"New protein synthesis is a prerequisite for maintaining long term memory," he said, "but you don't need this kind of transport forever, so it raises many questions that we want to answer. What molecules need to be synthesized to maintain memory? How long is this collection of RNAs stored? What localized mechanisms come into play for memory maintenance?"

###

In addition to Puthanveettil, who was the first author of the study, authors of "A Strategy to Capture and Characterize the Synaptic Transcriptome," include Igor Antonov, Sergey Kalchikov, Priyamvada Rajasethupathy, Yun-Beom Choi, Maxime Kinet, Irina Morozova, James J. Obama: Flight delay fix a 'Band-Aid'

A United Airlines jet departs in view of the air traffic control tower at Seattle-Tacoma International Airport Tuesday, April 23, 2013, in Seattle. A day after flight delays plagued much of the U.S., air travel is smoother Tuesday. But the government is warning passengers that the situation can change by the hour as it runs the nation's air traffic control system with a smaller staff. Airlines and members of Congress urged the Federal Aviation Administration to find other ways to make mandatory budget cuts besides furloughing controllers. While delays haven't been terrible yet, the airlines are worried about the long-term impact late flights will have on their budgets and on fliers. (AP Photo/Elaine Thompson)

(AP Photo/J. Scott Applewhite)

WASHINGTON (AP) ? President Barack Obama says the congressional fix for widespread flight delays is an irresponsible way to govern, but he's prepared to sign the legislation that lawmakers fast-tracked.

He says the bipartisan bill to end furloughs of air traffic controllers is a "Band-Aid" solution rather than a lasting answer to this year's $85 billion in across-the-board spending cuts known as the sequester.

The cuts have affected all federal agencies, and flight delays last week left thousands of travelers frustrated and furious and Congress feeling pressured to respond.

The Federal Aviation Administration announced Saturday that it had suspended all employee furloughs and that air traffic facilities would begin returning to regular staffing levels over the next 24 hours.

The FAA's statement said the air traffic system would resume normal operations by Sunday evening.

"Republicans claimed victory when the sequester first took effect, and now they've decided it was a bad idea all along," Obama said in his weekly radio and Internet address, aired Saturday.

He singled out the GOP even though the bill passed with overwhelming Democratic support in both the House and Senate.

The president scolded lawmakers for helping the Federal Aviation Administration while doing nothing to replace other cuts that he said harm federal employees, unemployed workers and preschoolers in Head Start.

"Maybe because they fly home each weekend, the members of Congress who insisted these cuts take hold finally realized that they actually apply to them, too," Obama said.

Rushed through Congress with remarkable speed, the bill marked a shift for Democrats who had hoped the impact of the cuts would increase pressure on Republicans to reverse the broad cuts.

Republicans have rejected Obama's proposal to replace the spending reductions with a mix of spending cuts and tax increases.

"There are some in the Obama administration who thought inflicting pain on the public would give the president more leverage to avoid making necessary spending cuts, and to impose more tax hikes on the American people," said Rep. Bill Shuster of Pennsylvania in the Republican address.

Shuster, chairman of the House Transportation and Infrastructure Committee, said the FAA could have averted the flight delays on its own by cutting costs elsewhere and rejiggering work schedules, but chose not to do so.

The bill signed by Obama would let the FAA use up to $253 million from an airport improvement program and other accounts to halt the furloughs through the Sept. 30 end of the government's fiscal year.

Faced with the prospect that emboldened Republicans will push to selectively undo other painful effects of the cuts, the White House said Friday that a piecemeal approach would be impractical, but wouldn't definitely rule out signing other fixes.

Liver Hormone Offers Hope for Diabetes Treatment

Betatrophin

Biologists have found a hormone in the liver that spurs the growth of insulin-secreting cells in the pancreas, a discovery they hope will lead to new treatments for diabetes.

A team led by Douglas Melton, co-director of the Harvard Stem Cell Institute in Cambridge, Massachusetts, identified the hormone, betatrophin, by inducing insulin resistance in mice using a peptide that binds to insulin receptors. That caused the animals' insulin-secreting pancreatic ? cells to proliferate. The researchers then searched for genes that showed increased activity, zeroing in on one that they were able to link to betatrophin production.?

Further experiments showed that 8-week-old mice injected with betatrophin showed an average 17-fold rise in the replication of their insulin-secreting pancreatic ? cells, the researchers report in Cell. Betatrophin is also found in the human liver, the team says.

?It?s rare that one discovers a new hormone, and this one is interesting because it?s so specific,? says Melton. ?It works only on ? cells?and it?s so robust and so potent.?

Pancreatic ? cells replicate rapidly during embryonic and neonatal stages in both mice and humans, but their growth falls off dramatically in adults. A decrease in the function of the cells late in life is the main cause of type 2 diabetes, a metabolic disorder that affects more than 300 million people worldwide. In the United States alone, the two forms of diabetes ? type 2 and type 1, which is caused by an autoimmune attack on pancreatic ? cells ? account for US$176 billion in direct medical costs each year.

Treatment hope
Melton thinks that injections of betatrophin once a month, or perhaps even once a year, could induce enough activity in pancreatic ? cells to provide the same level of blood-sugar regulation for people with type 2 diabetes as daily insulin injections do. But more importantly, he adds, they would cause fewer complications because the body would be making its own insulin. He also hopes that betatrophin will be able to help people with type 1 diabetes.

Matthias Hebrok, director of the University of California, San Francisco, Diabetes Center, says that the work ?is a great advance?. ?The findings are very interesting,? he says, although he would like to see the experiments repeated in older mice. ?Do mice that are on their way to becoming diabetic at an advanced age truly have an increase in proliferative capacity upon treatment with betatrophin?? he asks.

Henrik Semb, managing director of the Danish Stem Cell Center in Copenhagen, says that "the identification of a factor, betatrophin, that stimulates mouse ?-cell replication with remarkable efficiency is a very important discovery, because it provides the starting point for further studies to elucidate the underlying mechanism of ?-cell replication."

?-cell replication has proved difficult to control in humans.

Producing enough betatrophin for testing in human clinical trials will take about two years, according to Melton, who is also working to identify the hormone?s receptor and its mechanism of action.

The microbes you inhale on the New York City subway

Thursday, April 25, 2013

The microbial population in the air of the New York City subway system is nearly identical to that of ambient air on the city streets. This research, published ahead of print in the journal Applied and Environmental Microbiology, establishes an important baseline, should it become necessary to monitor the subway's air for dispersal of potentially dangerous microbes. Also, the combination of new methodologies in the study, including fast collection of aerosols and rapid sequencing technology, provide an efficient means for monitoring which was not previously available.

The results "are strong testimony for the efficiency of the train pumping system for ventilation," says principal investigator Norman R. Pace of the University of Colorado, Boulder. The wind one feels while walking across a subway grate as the subway clatters beneath also demonstrates just how effective that system is, he says. The only obvious differences in the subway's microbial population are the somewhat higher proportion of skin microbiota, and the doubled density of the fungal population, which Pace suggests may be due to rotting wood. "I was impressed by the similarity of [subway] and outdoor air," he says.

The researchers used a high tech mechanism to collect air at around 300 liters per minute (L/min), a big jump on the previous state of the art, which swallowed 12 L/min. That enabled collecting sufficient volume of air?a couple of cubic meters?to take the bacterial census within 20 minutes, instead of after "hours," says Pace. And analysis by sequencing is far faster and more thorough then using culture.

Pace notes that until now, the microbial content of subway air was unknown, and that the microbiology of indoor air is an emerging field of scientific inquiry. His research was funded by the Alfred P. Sloan Foundation, through its Microbiology of the Built Environment program, which has made 64 grants totaling $28 million to date.

"While it is difficult to predict what will be discovered on the frontier of scientific inquiry, the opportunity exists to better understand these complex microbial ecosystems and how they affect health and the environment. We expect that someday this knowledge will influence design and construction practices and other industrial processes," says Paula Olsiewski, program director, Alfred P. Sloan Foundation.

DEMANDING DINERS

Lively European birds called great tits haven?t kept up with climate changes that would require earlier nesting. Researchers have uncovered a quirk of population dynamics that?s giving the mistimed population some temporary protection.

Though climate change has knocked little birds called great tits out of sync with their chicks? food supply, the birds are maintaining their population numbers, a new study finds. But the way the tits cope may give them only a temporary reprieve.

?It?s buying them time,? says ecologist Tom Reed of the Netherlands Institute of Ecology in Wageningen.

Warmer springs mean that the peak food demands for Parus major chicks come later than the annual burst of local caterpillars. With less food, fewer young survive to leave their nests, Reed reports. However, the smaller resulting cohort of fledglings faces less competition for winter nuts and other resources. So a greater percentage of the youngsters survive through the winter than previously did, and ? so far ? this survival uptick has kept the study population from shrinking, Reed and an international team report in the April 26 Science.

These ?quite acrobatic little birds? are close relatives of North America?s chickadees, Reed says. Decades ago, researchers set out nest boxes in a Dutch national park, and great tits readily moved in, forming a manageable study population.

Intense monitoring allowed Marcel Visser, also at Wageningen and a coauthor of the study, to realize that the nest timing was going wrong. In 1998, he and a colleague reported that the park?s great tits wereone of the first documented examples of birds getting ecologically out of sync with food because of climate change.

Springtime in northern Europe had become unusually warm by the 1980s, and the trend has continued. The birds don?t migrate so they and the insects are exposed to the same weather. But caterpillars are more sensitive to a change in temperature, Reed says. The caterpillars and birds also take their temperature cues at different times in the spring. The differences mean the caterpillars tend to peak a lot earlier. The birds, unfortunately, haven?t kept pace.

With 37 years of data in the new study, researchers also detected that this mismatch is changing evolutionary pressures on the great tits. The breeding pairs that nest a bit earlier now produce more surviving offspring, so the population is getting a little nudge toward the new timing regimen. It remains to be seen whether the climate will change too fast for this slow adaptation to protect the population.

This kind of climate-induced mismatch between animals and their food supply has become a common worry among biologists. Pied flycatchers, small birds that spend winters in Africa, don?t return to Europe soon enough to catch the spring insect bonanza. In West Greenland,, migratory caribou miss the best grazing. And in Norway, puffins are declining as the spring bloom of plankton often misses the main hatching of herring and shrinks the -food supply later needed to feed puffin chicks.

Still unclear is whether any out-of-sync species besides the great tits would get some relief from the benefits of lessened competition among youngsters. ?We should be very cautious about suggesting that this is a widespread response to [food-supply] mismatch,? says ecologist Sarah Burthe from the Centre for Ecology and Hydrology in Edinburgh. Many studies, she says, already show that animals raise fewer total offspring in years with significant mismatches.

Samsung reportedly plans new 8inch tablet for June, Galaxy S 4 Active for July

Samsung Gulf president Young Soo Kim reportedly made casual mention of an upcoming rugged take on the Galaxy S 4 at a press event earlier in the week, but left out any details. Sources for the Wall Street Journal claim to know more of the story, and it sounds like the toughened phone won't be alone this summer. The Galaxy S 4 Active, as it's rumored to be called, would arrive in July and carry the dust- and waterproofing of spiritual predecessors like the Galaxy Xcover 2 while maintaining the GS 4's design language. A smaller, 4.3-inch parallel to the GS 4 would appear at the same time -- Galaxy S 4 Mini, anyone? The same tipsters also expect the release of a new 8-inch "Galaxy-line tablet" in June, which suggests the Galaxy Note 8.0 may get a regular Tab counterpart. We wouldn't base our buying strategies around the rumors when Samsung hasn't said anything that's truly on the record, but it might be wise to hold off if you're not in a rush.

'Redshirting' kindergarteners not as common as reported

Apr. 25, 2013 ? New research findings from the University of Virginia's Curry School of Education and the Stanford School of Education show that "redshirting" in kindergarten -- the practice of delaying for a year a child's entry into kindergarten -- is not happening at the rate previously reported.

In "'Academic Redshirting' in Kindergarten: Prevalence, Patterns, and Implications," published April 16 in the journal Educational Evaluation and Policy Analysis, study co-authors Daphna Bassok, assistant professor at the Curry School of Education, and Sean F. Reardon of Stanford found that only between 4 percent and 5.5 percent of children have their entry into kindergarten delayed.

Recent articles in popular news outlets and magazines have suggested that redshirting is more popular than it actually is. For instance, a 2008 New York Times article noted 17 percent of kindergartens were 6 years old rather than 5 when they started kindergarten.

"The perception recently has been that redshirting children for kindergarten is an increasingly popular practice, reaching upwards of nearly 20 percent of kindergarteners," Bassok said. "Our research shows that those numbers are way off and in reality we are seeing only about 4 percent of students starting kindergarten a year after they are first eligible."

Bassok noted that redshirting is garnering the attention of policymakers, in addition to education researchers.

The low rates nationwide, however, mask large variations across groups. According to the study, redshirting is extremely unlikely in schools in low-income neighborhoods, but in some high-income communities there are schools where nearly one in four children delay kindergarten entry.

Low-income and minority families are far more likely to indicate concerns about their child's readiness for kindergarten, a major reason to consider redshirting, but rarely delay kindergarten entry. While nearly 6 percent of white children are redshirted, the study found less than 1 percent of black children are; children from higher-income families redshirt almost three times as often as low-income children.

"Delaying kindergarten means finding and paying for another year of child care," Bassok said. "For most low-income families, redshirting is far too expensive."

The authors also find that redshirting is twice as likely to occur among boys than girls. One common explanation for redshirting is that children -- particularly boys -- are not prepared for the increasingly demanding kindergarten environment. However, in their study, Bassok and Reardon find that children who delay kindergarten do not seem to lag physically, socially or cognitively behind other children their age.

"We examined whether the children who delay kindergarten are those who seem to be struggling or immature, as evaluated by their preschool teachers, parents or even direct test scores," Bassok said. "We were surprised that at age 4, kids who end up delaying kindergarten looked just as 'ready' for school as their peers."

The study's findings suggest that redshirting parents are mostly concerned about their child's position relative to the other children in their classes. In communities where redshirting is common, there can be a large gap between the oldest and youngest students in a class. Parents whose children have birthdays close to the cut-off date for enrollment often would rather wait a year to enroll, so their child becomes among the oldest rather than youngest child in the group.

Bassok's current research is looking at whether or not the perception is true that the academic rigor of kindergarten is increasing, which she hopes will add more insight into the practice of redshirting.

South African aviator dies

JOHANNESBURG (AP) ? Laurie Kay, a South African pilot best known for flying a Boeing 747 passenger jet low over a Johannesburg stadium before the final of the 1995 Rugby World Cup, has died at the age of 67.

Kay had a suspected heart attack on Wednesday at the offices of the anti-rhinoceros poaching unit in Kruger National Park, South Africa's showcase game reserve, according to the country's parks service. Kay flew helicopters on patrols aimed at stopping poachers and was also doing technical work on a new anti-poaching surveillance aircraft, said Ike Phaahla, a parks spokesman.

"He was a great aviator," Phaahla said. "He was a gentleman. He had this outgoing personality. He made you feel very easy around him."

Kay was a South African Airways pilot when he swooped twice over Ellis Park stadium in a big jet at the start of the June 24, 1995, rugby final in which South Africa defeated New Zealand by a score of 15-12. The stenciled underside of the plane read "Good Luck Bokke" in a message of support for South Africa's national team, the Springboks.

One witness was Des Chetty, who was then a guard on the protective detail for F.W. de Klerk, the last president of the apartheid era that ended in 1994.

"It was a surreal feeling," Chetty wrote in an email. "One felt that if standing on the roof of the grandstands, you could have touched the belly of the plane. That's how close it felt. It was absolutely loud."

The daring stunt, depicted in the Hollywood movie "Invictus," electrified tens of thousands of fans who had no inkling that authorities had planned for a giant passenger jet to roar overhead. South Africa's ensuing victory on the field was a euphoric, unifying occasion for a country that had recently emerged from white racist rule.

Nelson Mandela, the anti-apartheid leader who became South Africa's first black president in 1994 elections, attended the game in a South African team shirt. It was a powerful gesture of reconciliation because rugby was a cultural bastion of white Afrikaners, who were guardians of the apartheid system.

Kay's flyover in a densely populated city flouted the most basic guidelines in international aviation, said John Carlin, author of "Playing the Enemy: Nelson Mandela and the Game That Made a Nation."

"The idea was to release maximum vibrations, noise and power into the stadium with a view to energizing the crowd and energizing the players," Carlin said in a telephone interview from Spain. "He got the plane very near to stalling in order to get that maximum power effect when he was over the stadium. It was pretty outrageous."

According to South African Airways, Kay and his crew had spent the previous week plotting the course, flying it many times in a light plane, and spending hours in a flight simulator. Director Clint Eastwood included the flyover episode in "Invictus," the 2009 film about South Africa's rugby victory that starred Morgan Freeman as Mandela.

The Boeing 747 was empty save for a small crew during the flyover, making it highly maneuverable despite its large size, noted Tony Smit, a longtime friend and pilot.

"There was nothing daredevil about Laurie," said Smit, who praised Kay for charity work and the mentoring of young pilots. "Everything that he did in aerobatics shows, everything is calculated to the finest degree."

Carlin said Kay, like many South African whites, initially had reservations about Mandela as the African National Congress leader negotiated an end to apartheid in the early 1990s. But that changed when the pair met on an international flight, according to the author. Mandela asked Kay, the captain, if he could upgrade members of his delegation from economy class, and Kay was charmed by Mandela's gracious manner.

"He was one more in a long, long line of people who succumbed to Mandela's charisma and general seductive abilities," Carlin said.

Even Mandela was startled during the 1995 flyover, said Carlin, who interviewed some of the president's bodyguards afterward.

"I don't think he had been prepared for this particular detail of the day's choreography," he said.

BlackBerry set to add Skype to its app line-up

TORONTO (Reuters) - BlackBerry said on Wednesday it plans to fill one of the biggest gaps in the app line-up on its new BlackBerry 10 devices with a long-awaited Skype application.

Skype, which was acquired by Microsoft Corp in 2011, allows its users to communicate via voice, video or chat over the Internet, by-passing traditional telephone networks. The service has hundreds of millions of users across the globe.

The announcement comes a day after BlackBerry said its new Q10 smartphone will be available in Canada on May 1, and in the United States before the end of May. The Q10, which comes with the physical keyboard that many of BlackBerry's core fan base cherish, is the second device to be powered by the company's new BB10 operating system.

Skype will be available on the Q10 on launch day. Users of the existing Z10 touchscreen device will have to wait a few weeks more to get Skype, after a software upgrade on the operating system, BlackBerry said.

A smaller app base than Apple's iOS and Google Inc's Android platforms has been one of the biggest criticisms of the new BlackBerry system. It has yet to offer some big-name apps like Netflix and Instagram.

Foursquare Redesigns Its Venue Pages For The Web To Capitalize On Its 50M Monthly Unique Visitors

Screenshot_4_22_13_8_44_PMFoursquare today launched redesigned venue pages to focus on its Explore and Discover functionality, bringing them more in line?with its homepage.?The changes, which come two weeks after the company raised another round of funding and released an updated iOS app, are designed to capitalize on the traffic that Foursquare gets from?Google, which has doubled the site’s traffic over the past year. This is an important play for Foursquare, as it’s competing with Google Local, Yelp and?Facebook,?which today launched its redesign for local business pages. Foursquare’s lead engineer for the web, Mike Singleton, told me that the site now gets over 50 million unique visitors on the web, which is 17 million more than actually use its app. That means that Foursquare is quietly breaking through as a place for information about venues, its most prized asset: People are coming from Google for different reasons, we needed to give them the information they needed at a glance, which was difficult. Since focusing on its Explore functionality, Singleton says that its usage has doubled, especially on the website. The new venue page has all of the information that people need in a quick glance, the pages are more visually appealing, and owners of the venues should be proud to show them off, perhaps by linking to them on their website over competitors like Yelp. Here’s a look at what a venue page looked like before today’s launch: You’ll notice that some of the more attractive content, such as photos, are pushed way down, putting more focus on the map. Additionally, information such as when the venue is open was shoved to the right-hand side, requiring a scroll, as well as someone’s eyes actually finding them. This redesign solves that: The new design is cleaner, and brings all of the information that Google searchers would want to see immediately, such as photos of the venue and a more attractive map. Since some of the people visiting from the web might not be app users, Foursquare wants to give them a better first impression of the service by populating the page with the info that’s needed to make quick decisions, such as whether to actually go to a place for dinner or not. By being able to search through tips, you could find information about a particular meal, rather than continually clicking “next” buttons to scan all tips. Naturally, Foursquare wants this page to

T-Mobile Unlimited Plans may get tethering bundled starting Wednesday

T-Mobile leaked screenshot

Value and Classic plans could get tethering pre-packaged with 500MB, 2.5GB, and 4.5GB options

In a screenshot recently obtained by TmoNews, it appears T-Mobile may be doing further restructuring of its post-paid plans. If true, unlimited data for both the Value and Classic plans will come pre-bundled with various tethering options starting on Wednesday. Basically, instead of buying unlimited data and a separate tethering bucket, subscribers will buy unlimited data with 500MB, 2.5GB, or 4.5GB of included tethering.

Currently, 500MB of tethering is already included with unlimited data. Instead of subscribers getting a $10 add-on for 2.5GB of tethering with their $20 unlimited data plan, the same plan will start at $30 and include tethering. For $40 a month, there is also a 4.5GB tethering option. Classic Plans will get the same changes: a $30 base plan with 500MB of tethering, a $40 plan with 2.5GB of tethering, and a $50 plan with 4.5GB of tethering.

It makes sense to see further simplification of T-Mobile's plans, making customer choices easier and less confusing (and in some cases cheaper). We'll be waiting for the official announcement in the coming days to see if these changes pan out.
